"Nanoscopium nominare libuit": far-field super resolution optical microscopy

Super-resolution fluorescence microscopy for biological research: from single cells to tissues and organs.

Several methodologies have been developed over the past several years for super-resolution fluorescence microscopy confirming the analysis done in the 50s. Among the most popular implementations, here we focus on stimulated emission depletion microscopy (STED), photoactivated localization microscopy (PALM), fluorescence photo activation localization microscopy (FPALM), and stochastic optical reconstruction microscopy (STORM).

Such methods have shown great promise for biological research even if having some individual strengths and weaknesses. Some recently developed original approaches aiming to 3D imaging of large scattering objects, pushing super resolution from single cells to tissues and organs, will be discussed. As well, application of super resolution methods to direct laser writing and coupling with atomic and force spectroscopy will be outlined.

The lecture will describe the basic principles for achieving super resolution using different physical approaches, demonstrate some applications in biology, and provide an overview of technical considerations for implementing and improving super resolution methods.
